The psychedelic drug in magic mushrooms may have lasting medical and spiritual benefits, according to new research from Johns Hopkins School of Medicine.
The mushroom-derived hallucinogen, called psilocybin, is known to trigger transformative spiritual states, but at high doses it can also result in "bad trips" marked by terror and panic. The trick is to get the dose just right, which the Johns Hopkins researchers report having accomplished.
In their study, the Hopkins scientists were able to reliably induce transcendental experiences in volunteers, which offered long-lasting psychological growth and helped people find peace in their lives — without the negative effects.
"The important point here is that we found the sweet spot where we can optimize the positive persistent effects and avoid some of the fear and anxiety that can occur and can be quite disruptive," says lead author Roland Griffiths, professor of behavioral biology at Hopkins.
Giffiths' study involved 18 healthy adults, average age 46, who participated in five eight-hour drug sessions with either psilocybin — at varying doses — or placebo. Nearly all the volunteers were college graduates and 78% participated regularly in religious activities; all were interested in spiritual experience.
Fourteen months after participating in the study, 94% of those who received the drug said the experiment was one of the top five most meaningful experiences of their lives; 39% said it was the single most meaningful experience.
Critically, however, the participants themselves were not the only ones who saw the benefit from the insights they gained: their friends, family member and colleagues also reported that the psilocybin experience had made the participants calmer, happier and kinder.
Ultimately, Griffiths and his colleagues want to see if the same kind of psychedelic experience could help ease anxiety and fear over the long term in cancer patients or others facing death. And following up on tantalizing clues from early research on hallucinogenic drugs like LSD, mescaline and psilocybin in the 1960s (which are all now illegal), researchers are also studying whether transcendental experiences could help spur recovery from addiction and treat other psychological problems like depression and post-traumatic stress disorder.
For Griffiths' current experiment, participants were housed in a living room-like setting designed to be calm, comfortable and attractive. While under the influence, they listened to classical music on headphones, wore eyeshades and were instructed to "direct their attention inward."
Each participant was accompanied by two other research-team members: a "monitor" and an "assistant monitor," who both had previous experience with people on psychedelic drugs and were empathetic and supportive. Before the drug sessions, the volunteers became acquainted enough with their team so that they felt familiar and safe. Although the experiments took place in the Hopkins hospital complex in order to ensure prompt medical attention in the event that it was needed, it never was.
As described by early advocates of the use of psychedelics — from ancient shamans to Timothy Leary and the Grateful Dead — the psilocybin experience typically involves a sense of oneness with the universe and with others, a feeling of transcending time, space and other limitations, coupled with a sense of holiness and sacredness. Overwhelmingly, these experiences are difficult to put into words, but many of Griffiths' participants said they were left with the sense that they understood themselves and others better and therefore had greater compassion and patience.

The library announced Thursday that it purchased 335 boxes of papers, videotapes, photographs and other items from the estate of Leary, who died in 1996.
Leary was a Harvard professor who became notorious for advocating the therapeutic use of psychedelic drugs. President Richard Nixon called Leary "the most dangerous man in America."
A trustee of Leary's estate tells The New York Times that the library paid $900,000 for the collection. Some of the money will be donated back to pay for the processing of the materials.
The collection includes letters from figures such as Allen Ginsberg, Ken Kesey, G. Gordon Liddy and Cary Grant.

As a retired clinical psychologist, Clark Martin was well acquainted with traditional treatments for depression, but his own case seemed untreatable as he struggled through chemotherapy and other grueling regimens for kidney cancer. Counseling seemed futile to him. So did the antidepressant pills he tried.

Nothing had any lasting effect until, at the age of 65, he had his first psychedelic experience. He left his home in Vancouver, Wash., to take part in an experiment at Johns Hopkins medical school involving psilocybin, the psychoactive ingredient found in certain mushrooms.
Scientists are taking a new look at hallucinogens, which became taboo among regulators after enthusiasts like Timothy Leary promoted them in the 1960s with the slogan “Turn on, tune in, drop out.” Now, using rigorous protocols and safeguards, scientists have won permission to study once again the drugs’ potential for treating mental problems and illuminating the nature of consciousness.
After taking the hallucinogen, Dr. Martin put on an eye mask and headphones, and lay on a couch listening to classical music as he contemplated the universe.
“All of a sudden, everything familiar started evaporating,” he recalled. “Imagine you fall off a boat out in the open ocean, and you turn around, and the boat is gone. And then the water’s gone. And then you’re gone.”
Today, more than a year later, Dr. Martin credits that six-hour experience with helping him overcome his depression and profoundly transforming his relationships with his daughter and friends. He ranks it among the most meaningful events of his life, which makes him a fairly typical member of a growing club of experimental subjects.
Researchers from around the world are gathering this week in San Jose, Calif., for the largest conference on psychedelic science held in the United States in four decades. They plan to discuss studies of psilocybin and other psychedelics for treating depression in cancer patients, obsessive-compulsive disorder, end-of-life anxiety, post-traumatic stress disorder and addiction to drugs or alcohol.
The results so far are encouraging but also preliminary, and researchers caution against reading too much into these small-scale studies. They do not want to repeat the mistakes of the 1960s, when some scientists-turned-evangelists exaggerated their understanding of the drugs’ risks and benefits.
Because reactions to hallucinogens can vary so much depending on the setting, experimenters and review boards have developed guidelines to set up a comfortable environment with expert monitors in the room to deal with adverse reactions. They have established standard protocols so that the drugs’ effects can be gauged more accurately, and they have also directly observed the drugs’ effects by scanning the brains of people under the influence of hallucinogens.
Scientists are especially intrigued by the similarities between hallucinogenic experiences and the life-changing revelations reported throughout history by religious mystics and those who meditate. These similarities have been identified in neural imaging studies conducted by Swiss researchers and in experiments led by Roland Griffiths, a professor of behavioral biology at Johns Hopkins.
In one of Dr. Griffiths’s first studies, involving 36 people with no serious physical or emotional problems, he and colleagues found that psilocybin could induce what the experimental subjects described as a profound spiritual experience with lasting positive effects for most of them. None had had any previous experience with hallucinogens, and none were even sure what drug was being administered.
To make the experiment double-blind, neither the subjects nor the two experts monitoring them knew whether the subjects were receiving a placebo, psilocybin or another drug like Ritalin, nicotine, caffeine or an amphetamine. Although veterans of the ’60s psychedelic culture may have a hard time believing it, Dr. Griffiths said that even the monitors sometimes could not tell from the reactions whether the person had taken psilocybin or Ritalin.
The monitors sometimes had to console people through periods of anxiety, Dr. Griffiths said, but these were generally short-lived, and none of the people reported any serious negative effects. In a survey conducted two months later, the people who received psilocybin reported significantly more improvements in their general feelings and behavior than did the members of the control group.
The findings were repeated in another follow-up survey, taken 14 months after the experiment. At that point most of the psilocybin subjects once again expressed more satisfaction with their lives and rated the experience as one of the five most meaningful events of their lives.
Since that study, which was published in 2008, Dr. Griffiths and his colleagues have gone on to give psilocybin to people dealing with cancer and depression, like Dr. Martin, the retired psychologist from Vancouver. Dr. Martin’s experience is fairly typical, Dr. Griffiths said: an improved outlook on life after an experience in which the boundaries between the self and others disappear.
In interviews, Dr. Martin and other subjects described their egos and bodies vanishing as they felt part of some larger state of consciousness in which their personal worries and insecurities vanished. They found themselves reviewing past relationships with lovers and relatives with a new sense of empathy.
“It was a whole personality shift for me,” Dr. Martin said. “I wasn’t any longer attached to my performance and trying to control things. I could see that the really good things in life will happen if you just show up and share your natural enthusiasms with people. You have a feeling of attunement with other people.”
The subjects’ reports mirrored so closely the accounts of religious mystical experiences, Dr. Griffiths said, that it seems likely the human brain is wired to undergo these “unitive” experiences, perhaps because of some evolutionary advantage.
“This feeling that we’re all in it together may have benefited communities by encouraging reciprocal generosity,” Dr. Griffiths said. “On the other hand, universal love isn’t always adaptive, either.”
Although federal regulators have resumed granting approval for controlled experiments with psychedelics, there has been little public money granted for the research, which is being conducted at Hopkins, the University of Arizona; Harvard; New York University; the University of California, Los Angeles; and other places.
The work has been supported by nonprofit groups like the Heffter Research Institute and MAPS, the Multidisciplinary Association for Psychedelic Studies.
“There’s this coming together of science and spirituality,” said Rick Doblin, the executive director of MAPS. “We’re hoping that the mainstream and the psychedelic community can meet in the middle and avoid another culture war. Thanks to changes over the last 40 years in the social acceptance of the hospice movement and yoga and meditation, our culture is much more receptive now, and we’re showing that these drugs can provide benefits that current treatments can’t.”
Researchers are reporting preliminary success in using psilocybin to ease the anxiety of patients with terminal illnesses. Dr. Charles S. Grob, a psychiatrist who is involved in an experiment at U.C.L.A., describes it as “existential medicine” that helps dying people overcome fear, panic and depression.
“Under the influences of hallucinogens,” Dr. Grob writes, “individuals transcend their primary identification with their bodies and experience ego-free states before the time of their actual physical demise, and return with a new perspective and profound acceptance of the life constant: change.”

Albert Hofmann, the father of the mind-altering drug LSD whose medical discovery inspired — and arguably corrupted — millions in the 1960s hippie generation, has died. He was 102.
Hofmann died Tuesday at his home in Burg im Leimental, said Doris Stuker, a municipal clerk in the village near Basel where Hofmann moved following his retirement in 1971.
For decades after LSD was banned in the late 1960s, Hofmann defended his invention.
"I produced the substance as a medicine. ... It's not my fault if people abused it," he once said.
The Swiss chemist discovered lysergic acid diethylamide-25 in 1938 while studying the medicinal uses of a fungus found on wheat and other grains at the Sandoz pharmaceuticals firm in Basel.
He became the first human guinea pig of the drug when a tiny amount of the substance seeped onto his finger during a repeat of the laboratory experiment on April 16, 1943.
"I had to leave work for home because I was suddenly hit by a sudden feeling of unease and mild dizziness," he subsequently wrote in a memo to company bosses.
"Everything I saw was distorted as in a warped mirror," he said, describing his bicycle ride home. "I had the impression I was rooted to the spot. But my assistant told me we were actually going very fast."
Upon reaching home, Hofmann sat down on a divan and began experiencing what he called "wonderful visions."
"What I was thinking appeared in colors and in pictures," he told Swiss television network SF DRS for a program marking his 100th birthday two years ago. "It lasted for a couple of hours and then it disappeared."
Three days later, Hofmann experimented with a larger dose. The result was the world's first scientifically documented bad trip.
"The substance which I wanted to experiment with took over me. I was filled with an overwhelming fear that I would go crazy. I was transported to a different world, a different time," Hofmann wrote.
Hofmann and his scientific colleagues hoped that LSD would make an important contribution to psychiatric research. The drug exaggerated inner problems and conflicts and thus it was hoped that it might be used to recognize and treat mental illnesses like schizophrenia.
For a time, Sandoz sold LSD 25 under the name Delysid, encouraging doctors to try it themselves. It was one of the strongest drugs in medicine — with just one gram enough to drug an estimated 10,000 to 20,000 people for 12 hours.
Hofmann discovered the drug had a similar chemical structure to psychedelic mushrooms and herbs used in religious ceremonies by Mexican Indians.
LSD was elevated to international fame in the late 1950s and 1960s thanks to Harvard professor Timothy Leary who embraced the drug under the slogan "turn on, tune in, drop out." The film star Cary Grant and numerous rock musicians extolled its virtues in achieving true self discovery and enlightenment.
But away from the psychedelic trips and flower children, horror stories emerged about people going on murder sprees or jumping out of windows while hallucinating. Heavy users suffered permanent psychological damage.
The U.S. government banned LSD in 1966 and other countries followed suit.
Hofmann maintained this was unfair, arguing that the drug was not addictive. He repeatedly argued for the ban to be lifted to allow LSD to be used in medical research.
He himself took the drug — purportedly on an occasional basis and out of scientific interest — for several decades.
"LSD can help open your eyes," he once said. "But there are other ways — meditation, dance, music, fasting."
Even so, the self described "father" of LSD readily agreed that the drug was dangerous if in the wrong hands. This was reflected by the title of his 1979 book: "LSD - my problem child."
In it he wrote that, "The history of LSD to date amply demonstrates the catastrophic consequences that can ensue when its profound effect is misjudged and the substance is mistaken for a pleasure drug."
Hofmann retired from Sandoz in 1971. He devoted his time to travel, writing and lectures — which often reflected his growing interest with philosophy and religious questions.
He remained active, going for walks in the small picturesque village where he lived in the Swiss Jura mountains, a stone's throw from the French border. He spoke at a Basel ceremony honoring him on his 100th birthday.
"This is really a high point in my advanced age," Hofmann said. "You could say it is a consciousness-raising experience without LSD."
Hofmann is survived by two of his four children. He was predeceased by his wife Anita.
Funeral arrangements were not immediately available.

"What I was thinking appeared in colors and in pictures," he told Swiss television network SF DRS for a program marking his 100th birthday two years ago. "It lasted for a couple of hours and then it disappeared."
Three days later, Hofmann experimented with a larger dose. The result was the world's first scientifically documented bad trip.
"The substance which I wanted to experiment with took over me. I was filled with an overwhelming fear that I would go crazy. I was transported to a different world, a different time," Hofmann wrote.
Hofmann and his scientific colleagues hoped that LSD would make an important contribution to psychiatric research. The drug exaggerated inner problems and conflicts and thus it was hoped that it might be used to recognize and treat mental illnesses like schizophrenia.
For a time, Sandoz sold LSD 25 under the name Delysid, encouraging doctors to try it themselves. It was one of the strongest drugs in medicine — with just one gram enough to drug an estimated 10,000 to 20,000 people for 12 hours.
Hofmann discovered the drug had a similar chemical structure to psychedelic mushrooms and herbs used in religious ceremonies by Mexican Indians.
LSD was elevated to international fame in the late 1950s and 1960s thanks to Harvard professor Timothy Leary who embraced the drug under the slogan "turn on, tune in, drop out." The film star Cary Grant and numerous rock musicians extolled its virtues in achieving true self discovery and enlightenment.
But away from the psychedelic trips and flower children, horror stories emerged about people going on murder sprees or jumping out of windows while hallucinating. Heavy users suffered permanent psychological damage.
The U.S. government banned LSD in 1966 and other countries followed suit.
Hofmann maintained this was unfair, arguing that the drug was not addictive. He repeatedly argued for the ban to be lifted to allow LSD to be used in medical research.
He himself took the drug — purportedly on an occasional basis and out of scientific interest — for several decades.
"LSD can help open your eyes," he once said. "But there are other ways — meditation, dance, music, fasting."
Even so, the self described "father" of LSD readily agreed that the drug was dangerous if in the wrong hands. This was reflected by the title of his 1979 book: "LSD - my problem child."
In it he wrote that, "The history of LSD to date amply demonstrates the catastrophic consequences that can ensue when its profound effect is misjudged and the substance is mistaken for a pleasure drug."
Hofmann retired from Sandoz in 1971. He devoted his time to travel, writing and lectures — which often reflected his growing interest with philosophy and religious questions.
He remained active, going for walks in the small picturesque village where he lived in the Swiss Jura mountains, a stone's throw from the French border. He spoke at a Basel ceremony honoring him on his 100th birthday.
"This is really a high point in my advanced age," Hofmann said. "You could say it is a consciousness-raising experience without LSD."
Hofmann is survived by two of his four children. He was predeceased by his wife Anita.
Funeral arrangements were not immediately available.
